{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2026,2,12]],"date-time":"2026-02-12T15:48:18Z","timestamp":1770911298491,"version":"3.50.1"},"reference-count":20,"publisher":"Wiley","issue":"9","license":[{"start":{"date-parts":[[2007,9,17]],"date-time":"2007-09-17T00:00:00Z","timestamp":1189987200000},"content-version":"vor","delay-in-days":0,"URL":"http:\/\/onlinelibrary.wiley.com\/termsAndConditions#vor"}],"content-domain":{"domain":[],"crossmark-restriction":false},"short-container-title":["Cancer"],"published-print":{"date-parts":[[2007,11]]},"abstract":"<jats:title>Abstract<\/jats:title><jats:sec><jats:title>BACKGROUND.<\/jats:title><jats:p>Methylation patterns may be useful biomarkers of cancer detection and risk assessment.<\/jats:p><\/jats:sec><jats:sec><jats:title>METHODS.<\/jats:title><jats:p>The methylation status of 6 genes, including a candidate tumor suppressor gene (<jats:italic>BLU<\/jats:italic>), the cadherin 13 gene (<jats:italic>CDH13<\/jats:italic>), the fragile histidine triad gene (<jats:italic>FHIT<\/jats:italic>), the cell cycle control gene <jats:italic>p16<\/jats:italic>, the retinoic acid receptor \u03b2 gene (<jats:italic>RAR<\/jats:italic>\u03b2), and the Ras association domain family 1 gene (<jats:italic>RASSF1A<\/jats:italic>), was examined in plasma samples, corresponding tumor tissues, and normal lung tissues from a group of 63 patients with lung cancer and in plasma samples from 36 cancer\u2010free individuals. The detection rate of the <jats:italic>p16<\/jats:italic> gene was validated in a test group of 20 patients with lung cancer.<\/jats:p><\/jats:sec><jats:sec><jats:title>RESULTS.<\/jats:title><jats:p>The concordance of methylation in tumor tissues and plasma samples was 86%, 87%, 80%, 75%, 76%, and 84% for the <jats:italic>BLU<\/jats:italic>, <jats:italic>CDH13<\/jats:italic>, <jats:italic>FHIT<\/jats:italic>, <jats:italic>p16<\/jats:italic>, <jats:italic>RAR<\/jats:italic>\u03b2, and <jats:italic>RASSF1A<\/jats:italic> genes, respectively. The test group showed a similar concordance for <jats:italic>p16<\/jats:italic> methylation detection. Multiple logistic regression analysis showed that the odds ratio for having lung cancer was 10.204 for individuals with <jats:italic>p16<\/jats:italic> methylation (<jats:italic>P<\/jats:italic> = .013) and 9.952 for individuals with <jats:italic>RASSFIA<\/jats:italic> methylation (<jats:italic>P<\/jats:italic> = .019). After several trial tests, the authors established that methylation for \u22652 of the 6 markers met the criterion for an elevated risk of cancer. Comparisons yielded a sensitivity of 73%, a specificity of 82%, and a concordance of 75% between the methylation patterns in tumor tissues and in corresponding plasma samples. The detection rate was relatively high in cigarette smokers with advanced squamous cell lung cancer.<\/jats:p><\/jats:sec><jats:sec><jats:title>CONCLUSIONS.<\/jats:title><jats:p>The current results indicated that multiple epigenetic markers in the plasma, especially the <jats:italic>p16<\/jats:italic> and <jats:italic>RASSF1A<\/jats:italic> genes, can be used for lung cancer detection. This methylation marker panel should improve the detection of cancer or the risk assessment for lung cancer in combination with conventional diagnostic tools.
