The idiotypic marker detected by this antiserum (the NP<jats:sup>b<\/jats:sup> idiotype) requires for its expression a specific heavy (H) chain\u2010lambda (\u039b) light (L) chain combination as shown in chain reassociation experiments. The idiotypic binding reaction is inhibitable by hapten. BALB\/c and CBA mice also produce large amounts of \u039b\u2010bearing anti\u2010NP antibodies, but these do not express the NP<jats:sup>b<\/jats:sup> idiotype as defined by the rabbit antiserum. Since the NP<jats:sup>b<\/jats:sup> idiotype could be reconstituted by reassociating C57 BL\/6 anti\u2010NP antibody H chains with either HOPC 1, or BALB\/c or CBA anti\u2010NP antibody \u039b chains, the absence of the NP<jats:sup>b<\/jats:sup> idiotype in BALB\/c and CBA antibodies implies that these animals express different sets of H chain variable (V) regions in the anti\u2010NP response.<\/jats:p><jats:p>SJL mice, in contrast to other mouse strains with the Ig\u20101<jats:sup>b<\/jats:sup> allotype, express only traces of NP<jats:sup>b<\/jats:sup> idiotype in their anti\u2010NP antibodies, the L chains of which are almost exclusively of the \u03c7 type.<\/jats:p><jats:p>Knowing that SJL mice possess a regulatory gene responsible for the low level of \u039b (\u039b<jats:sup>10<\/jats:sup>) chains in their immunoglobulins, we have analyzed the progeny of (SJL \u00d7 BALB\/C)F<jats:sub>1<\/jats:sub>, (SJL \u00d7 BALB\/C)F<jats:sub>1<\/jats:sub> \u00d7 BALB\/C and (SJL \u00d7 BALB\/C)F<jats:sub>1<\/jats:sub> \u00d7 SJL for the expression of \u039b\u2010bearing anti\u2010NP antibodies and NP<jats:sup>b<\/jats:sup> idiotype.<\/jats:p><jats:p>In accord with previous work (SJL \u00d7 BALB\/c)F<jats:sub>1<\/jats:sub> mice produced substantial amounts of anti\u2010NP antibodies with the NP<jats:sup>b<\/jats:sup> idiotype. Backcross of these mice to BALB\/c revealed linkage of the NP<jats:sup>b<\/jats:sup> idiotype to the Ig\u20101<jats:sup>b<\/jats:sup> allotype, in agreement with previous data. Backcross of the F<jats:sub>1<\/jats:sub> animals to SJL demonstrated control of the NP<jats:sup>b<\/jats:sup> idiotype by the gene regulating \u039b chain expression in serum immunoglobulin.<\/jats:p><jats:p>Chain reassociation experiments showed that in SJL mice, the regulation against \u039b chain expression prevents the selection in the anti\u2010NP response of those H chains that together with \u039b chains constitute the NP<jats:sup>b<\/jats:sup> idiotype.
