The hybrid cell lines obtained were analyzed for heavy and light chain distribution, fine specificity, and idiotype.<\/jats:p><jats:p>The majority of monoclonal antibodies possessed either \u03b3<jats:sup>1<\/jats:sup> or \u03bc chains. The distribution of L chains among these antibodies was approximately half \u03bb and half \u03f0. Thirteen monoclonal antibodies were grown as ascites tumors in mice. Examination of their fine specificity patterns showed that all of the \u03bb antibodies are heteroclitic and have similar fine specificity patterns. Five of the seven \u03f0 antibodies are also heteroclitic, but their fine specificity patterns are more heterogeneous than those of the \u03bb antibodies.<\/jats:p><jats:p>Polyspecific anti\u2010idiotypic sera directed against pooled primary serum antibody (R \u0101\u2010NP<jats:sup>a<\/jats:sup>) or against individual monoclonals were used for idiotypic characterization of the monoclonal antibodies. The R \u0101\u2010NP<jats:sup>a<\/jats:sup> bound all of the \u03bb antibodies but none of the \u03f0 antibodies suggesting that the \u03f0 antibodies may be much more heterogeneous and were therefor not recognized in the presence of the more homogeneous \u03bb antibodies. Further idiotypic analysis demonstrated that the \u03bb antibodies, although no two are identical, are a very homogeneous group of antibodies which cross\u2010react with one another but not with the \u03f0 antibodies. Some, but not all, of the \u03f0 antibodies cross\u2010react with each other although none are cross\u2010reactive with the \u03bb antibodies. Because the \u03bb\u2010associated idiotype is recognized by the R \u0101\u2010NP<jats:sup>a<\/jats:sup> and its characteristics are similar to that of the C57BL\/6 major idiotype (NP<jats:sup>b<\/jats:sup>), it is referred to as NP<jats:sup>a<\/jats:sup>.
