Supernatants of this hybrid line or its active clones as well as ascitic fluids, aspirated from the peritoneum of mice injected with the hybridoma cells, manifest a(T,G)\u2010A\u2010L\u2010specific helper T cell\u2010replacing activity as assayed in an <jats:italic>in vitro<\/jats:italic> hapten\u2010carrier antibody production system. The factor activity can be specifically absorbed on and eluted from columns of immobilized (T,G)\u2010A\u2013L, antibodies directed against the variable region of the immunoglobulin heavy chain and an antiserum directed against (T,G)\u2010A\u2010L\u2010specific idiotypes. Although the R\u20109 factor and anti\u2010(T,G)\u2010A\u2010L antibodies share V region gene\u2010encoded determinants, they differ in their fine antigenic specificity, as determined by cross\u2010reactivity with closely related synthetic polypeptide immunogens. Whereas anti\u2010(T,G)\u2010A\u2013L antibodies bind equally well to the L\u2010phenylalanine\u2010 or L\u2010histidine\u2010containing polypeptides (Phe,G)\u2010A\u2010L and (H,G)\u2010A\u2010L, to poly(LGlu)\u2010poly(DLAla)\u2010poly(LLys) (G\u2010A\u2013L) and poly(Tyr,Glu)\u2010poly(LPro)\u2010poly(LLys) [(T,G)\u2010Pro\u2013L], the R\u20109 factor was completely absorbed on columns of (Phe,G)\u2010A\u2010L and G\u2010A\u2010L and only partially absorbed on immobilized (T,G)\u2010Pro\u2013L and (H,G)\u2010A\u2010L. In addition to V region gene products, the R\u20109 factor also bears major histocompatibility complex (MHC) determinants, which were found to map to the left end of the I<jats:sup>b<\/jats:sup> region.
