Immunoprecipitations carried out with 6 different monoclonal antibodies (mAb) against LFA\u20101 indicated that four mAb (SPV\u2010 L1, SPV\u2010L5, SPV\u2010L7 and SPV\u2010L11) were directed against the a chain, whereas mAb CLB54 and MHM\u201023 were found to react with the common \u03b2 chain of LFA\u20101, Mo\u20101 and p150,95. LFA\u20101 and Mo\u20101 expressed on KG\u20101 cells or lymphocytes, monocytes and granulocytes from one donor were homogeneous. Interestingly the a chain of p150,95 showed heterogeneity. The molecular weight of the a chain expressed on monocytes was consistently higher than that of the \u03b1 chain on granulocytes. The \u03b2 subunits of LFA\u20101 and Mo\u20101 (as detected by mAb Bear\u20101) are not only similar in molecular weight and isoelectric focusing patterns, but it is demonstrated here that they are also identically glycosylated and have similar protein backbones as judged by tryptic peptide mapping. In spite of their structural similarities, LFA\u20101 and Mo\u20101 differ completely in some of their biological functions. Anti\u2010LFA\u20101 mAb strongly inhibited monocyte\u2010dependent T cell proliferation induced by tetanus toxoid or <jats:italic>Helix pomatia<\/jats:italic> hemocyanin and pokeweed mitogen\u2010driven specific antibody production <jats:italic>in vitro<\/jats:italic>, whereas the anti\u2010Mo\u20101 antibody Bear\u20101 was ineffective.
