We show here that adult murine thymus contains a subpopulation of CD4\u20108<jats:sup>+<\/jats:sup> cells that differ from CD4<jats:sup>\u2212<\/jats:sup>8<jats:sup>+<\/jats:sup> cells found d the periphery in that they do not express the T cell receptor\u2010associated CD3 molecular complex. Such CD3<jats:sup>\u2212<\/jats:sup>4<jats:sup>\u2212<\/jats:sup>8<jats:sup>+<\/jats:sup> thymocytes are cortisone sensitive and rapidly cycling <jats:italic>in situ.<\/jats:italic> Furthermore, in contrast to mature T cells, most CD3<jats:sup>\u2212<\/jats:sup>4<jats:sup>\u2212<\/jats:sup>8<jats:sup>+<\/jats:sup> thymocytes express low levels of CD5 and high levels of the B2A2 antigen. CD3<jats:sup>\u2212<\/jats:sup>4<jats:sup>\u2212<\/jats:sup>8<jats:sup>+<\/jats:sup> thymocytes fail to respond to a variety of mitogenic stimuli <jats:italic>in vitro<\/jats:italic> but do give rise upon short\u2010term culture to CD4<jats:sup>+<\/jats:sup>8<jats:sup>+<\/jats:sup> cells.
