Unexpectedly, we found that human mature B cells producing \u03c7 chains and having germ\u2010line \u03bb genes contain \u03bb mRNA consisting of an invariant 5\u2032 region (herein termed X) and of one of the classical C<jats:sub>\u03bb<\/jats:sub> exons. The X region of these transcripts originates from a unique exon located 5 kb upstream of the J\u2010C<jats:sub>\u03bb<\/jats:sub>1 gene segment. X\u2010C<jats:sub>\u03bb<\/jats:sub> mRNA expression occurs without somatic DNA rearrangement. The use of the X DNA fragment as a probe allows definition of a family of human genes that comprises at least four members and includes the first exon of the \u03bb<jats:sub>14.1<\/jats:sub> gene. The latter is selectively transcribed in pre\u2010B lymphocytes and directs the synthesis of a \u03bb\u2010like chain. In contrast, the X\u2010C<jats:sub>\u03bb<\/jats:sub> transcripts do not appear to encode a C<jats:sub>\u03bb<\/jats:sub>\u2010related polypeptide in mature B cells.
