We show that the mature form of the antigen is a heterodimer comprising a 150\u2010kDa \u03b1 chain and a 120\u2010kDa \u03b2 chain. Direct sequencing of tryptic peptides cleaved from the purified \u03b2 chain identified this polypeptide with the integrin \u03b27 isotype. cDNA clones coding for the \u03b27 chain have recently been isolated from T cell cDNA libraries, but the \u03b27 chain had not been identified at the protein level. No information is available concerning the primary structure of the HML\u20101 \u03b1 chain. We show that this subunit is synthesized as a precursor form that undergoes, like several other integrin \u03b1 subunits, a post\u2010translational cleavage of a peptide bond. Among the 11 human integrin \u03b1 chains previously identified, 10 have biochemical features and\/or a distribution different from those of HML\u20101 \u03b1. One, VLA\u03b14 (CD49d), has a molecular mass of 150 kDa and is expressed on HML\u20101<jats:sup>+<\/jats:sup> cells but is not recognized by HML\u20101 mAb. We conclude that HML\u20101 is a novel member of the integrin family made of the \u03b27 chain and of an as\u2010yet\u2010undescribed human \u03b1 chain characterized by the post\u2010translational cleavage of a 10\u2010kDa peptide.
