Characterization of the specificities of IgG<jats:sub>1<\/jats:sub> antibodies produced by single, sorted IgG<jats:sub>1<\/jats:sub><jats:sup>+<\/jats:sup> NP<jats:sup>+<\/jats:sup> cells in both Elispot assays and in microcultures containing lipo\u2010polysaccharide, interleukin (IL)\u20102, IL\u20104 and IL\u20105 indicates that the splenic IgG<jats:sub>1<\/jats:sub><jats:sup>+<\/jats:sup> NP<jats:sup>+<\/jats:sup> B cell population includes both IgG<jats:sub>1<\/jats:sub> anti\u2010NP antibody\u2010secreting cells and non\u2010secreting, IgG<jats:sub>1<\/jats:sub><jats:sup>+<\/jats:sup> memory B cells. Each functionally discrete population of IgG<jats:sub>1<\/jats:sub><jats:sup>+<\/jats:sup> B cells expresses a distinctive surface phenotype defined by a wide range of B cell markers. In particular, antibody\u2010secreting, IgG<jats:sub>1<\/jats:sub><jats:sup>+<\/jats:sup> cells were uniquely identified by co\u2010expression of the matrix receptor, syndecan. The NP\u2010specific B cell population emerging in the day 7 primary response was assessed for clonotypic diversity by amplification and direct sequencing of the rearranged V186.2 heavy chain variable region gene expressed by single, <jats:italic>ex vivo<\/jats:italic> IgG<jats:sub>1<\/jats:sub><jats:sup>+<\/jats:sup> NP<jats:sup>+<\/jats:sup> \u03bb<jats:sup>+<\/jats:sup> B cells. Memory B cell clones, distinguished by junctional diversity, carried either no mutation or a single mutation within rearranged V186.2, suggesting isolation of these cells at the onset of the hypermutation mechanism.
