In adult lymph nodes, \u03b1<jats:sub>IEL<\/jats:sub> was expressed at low levels by 40\u201370% of CD8<jats:sup>+<\/jats:sup> T cells and &lt; 5% of CD4<jats:sup>+<\/jats:sup> T cells. However, virtually all intestinal intraepithelial lymphocytes and \u02dc20% of lamina propria CD4<jats:sup>+<\/jats:sup> T cells were 2E7<jats:sup>+<\/jats:sup>, indicating a preferential expression of this integrin on mucosal T cells. Examination of \u03b1<jats:sub>IEL<\/jats:sub> integrin expression during thymus ontogeny revealed that \u02dc3\u20135% of fetal or adult thymocytes were 2E7<jats:sup>+<\/jats:sup>. Interestingly, early in fetal thymus ontogeny, \u02dc40% of 2E7<jats:sup>+<\/jats:sup> cells expressed T cell receptor (TcR)\u2010\u03b3\u03b4 and this subset persisted through birth. A developmental switch occurred such that 2E7<jats:sup>+<\/jats:sup> TcR<jats:sup>\u2212<\/jats:sup> CD4<jats:sup>\u2212<\/jats:sup>8<jats:sup>+<\/jats:sup> cells detected on fetal day 19 were followed by 2E7<jats:sup>+<\/jats:sup> TcR\u2010\u03b1\u03b2 CD4<jats:sup>\u2212<\/jats:sup>8<jats:sup>+<\/jats:sup> cells in the neonatal thymus. The latter population persisted throughout thymus ontogeny into adulthood. Interestingly, a subset of TcR\u2010\u03b3\u03b4 V\u03b33<jats:sup>+<\/jats:sup> day 16 fetal thymocyte dendritic epidermal cell (DEC) precursors were 2E7<jats:sup>+<\/jats:sup>, but all mature DEC expressed high levels of \u03b1<jats:sub>IEL<\/jats:sub> integrin, suggesting that the \u03b1<jats:sub>IEL<\/jats:sub> integrin was acquired late in DEC maturation. This possibility was strenghthened by immunohistochemical localization of the majority of 2E7<jats:sup>+<\/jats:sup> \u03b3\u03b4 and \u03b1\u03b2 T cells to the medullary regions of the thymus.
