As most inbred mouse strains and wild mice contain integrated MMTV DNA, knowing the precise extent of MMTV influence on T cell development is required in order to study T cell immunobiology in the mouse. In this report, backcross breeding between BALB.D2 (<jats:italic>Mtv\u20106, \u22127, \u22128<\/jats:italic> and \u2212<jats:italic>9<\/jats:italic>) and 38CH (<jats:italic>Mtv<\/jats:italic><jats:sup>\u2212<\/jats:sup>) mice was carried out to obtain animals either lacking endogenous MMTV or containing a single MMTV locus, <jats:italic>i.e. Mtv\u20106, \u22127, \u22128<\/jats:italic> or \u2212<jats:italic>9<\/jats:italic>. The TcR V\u03b2 chain (TcR V\u03b2) usage in these mice was analyzed using monoclonal antibodies specific for TcR V\u03b22,V\u03b23, V\u03b24,\u03b25,V\u03b26,V\u03b27,V\u03b28,V\u03b211,V\u03b212 and V\u03b214 segments. Both <jats:italic>Mtv\u20108<\/jats:italic><jats:sup>+<\/jats:sup> mice and <jats:italic>Mtv\u20109<\/jats:italic><jats:sup>+<\/jats:sup> mice deleted TcR V\u03b25<jats:sup>+<\/jats:sup> and V\u03b211<jats:sup>+<\/jats:sup> T cells. Moreover, we also observed the deletion of TcR V\u03b212<jats:sup>+<\/jats:sup> cells by <jats:italic>Mtv\u20108<\/jats:italic> and <jats:italic>Mtv\u20109<\/jats:italic> products. <jats:italic>Mtv\u20106<\/jats:italic><jats:sup>+<\/jats:sup> and <jats:italic>Mtv\u20107<\/jats:italic><jats:sup>+<\/jats:sup> animals deleted TcR V\u03b23<jats:sup>+<\/jats:sup> and V\u03b235<jats:sup>+<\/jats:sup> cells, and TcR V\u03b26<jats:sup>+<\/jats:sup>,V\u03b27<jats:sup>+<\/jats:sup> and V\u03b28.1<jats:sup>+<\/jats:sup> cells, respectively. Unexpectedly, TcR V\u03b28.2<jats:sup>+<\/jats:sup> cells were also deleted in some backcross mice expressing <jats:italic>Mtv\u20107<\/jats:italic>. TcR V\u03b28.2 reactivity to <jats:italic>Mtv\u20107<\/jats:italic> was shown to be brought by the 38CH strain and to result from an amino acid substitution (Asn \u2192 Asp) in position 19 on the TcR V\u03b28.2 fragment. Reactivities of BALB.D2 TcR V\u03b28.2 and 38CH TcR V\u03b28.2 to the exogenous infectious viruses, MMTV(SW) and MMTV(SHN), were compared.<\/jats:p><jats:p>Finally, the observation of increased frequencies of TcR V\u03b22<jats:sup>+<\/jats:sup>, V\u03b24<jats:sup>+<\/jats:sup> and V\u03b28<jats:sup>+<\/jats:sup> CD4<jats:sup>+<\/jats:sup> T cell subsets in <jats:italic>Mtv\u20108<\/jats:italic><jats:sup>+<\/jats:sup> and <jats:italic>Mtv\u20109<\/jats:italic><jats:sup>+<\/jats:sup> mice, and TcR V\u03b24<jats:sup>+<\/jats:sup> CD4<jats:sup>+<\/jats:sup> T cells in <jats:italic>Mtv\u20106<\/jats:italic><jats:sup>+<\/jats:sup> and <jats:italic>Mtv\u20107<\/jats:italic><jats:sup>+<\/jats:sup> mice, when compared with the T cell repertoire of <jats:italic>Mtv<\/jats:italic><jats:sup>\u2212<\/jats:sup> mice, is consistent with the possibility that MMTV products contribute to positive selection of T cells.<\/jats:p>","DOI":"10.1002\/eji.1830250334","type":"journal-article","created":{"date-parts":[[2007,3,2]],"date-time":"2007-03-02T00:25:04Z","timestamp":1172795104000},"page":"857-862","source":"Crossref","is-referenced-by-count":12,"title":["T cell receptor V\u03b2 repertoire in mice lacking endogenous mouse mammary tumor provirus"],"prefix":"10.1002","volume":"25","author":[{"given":"Michel Y.","family":"Braun","sequence":"first","affiliation":[]},{"given":"Evelyne","family":"Jouvin\u2010Marche","sequence":"additional","affiliation":[]},{"given":"Patrice N.","family":"Marche","sequence":"additional","affiliation":[]},{"given":"H.
