Efficient L<jats:sup>d<\/jats:sup>\u2010restricted presentation of the S<jats:sub>28\u201339<\/jats:sub> epitope to CTL is observed in cells of different tissue origin pulsed <jats:italic>in vitro<\/jats:italic>, either with the antigenic S<jats:sub>28\u201339<\/jats:sub> 12\u2010mer S\u2010peptide, or with particulate S\u2010antigen. The kinetics of epitope presentation differ in S\u2010peptide\u2010pulsed and in S\u2010particle\u2010pulsed cells: while a 15\u2010min pulse with the antigenic peptide sensitizes targets for class I\u2010restricted CTL lysis, presentation of S\u2010particles requires 30\u201360 min to sensitize cells for CTL lysis. Uptake of antigenic material and active metabolism of the presenting cell are required for processing of S\u2010particles, but not for sensitizing targets with S\u2010peptides. Intracellular processing and presentation of S\u2010particles is blocked in cells treated with chloroquine, NH<jats:sub>4<\/jats:sub>Cl, primaquine, or leupeptin, but not by treatment with cycloheximide or brefeldin A.
