Treatment with 12\u2010o\u2010<jats:italic>\u2010<\/jats:italic>tetradecanoyl phorbol 13\u2010acetate (TPA), inducing macrophage\u2010like differentiation, produces a dramatic decrease of MHC\u2010II expression as result of down\u2010modulation of the activation of immune response gene\u20041 (<jats:italic>AIR\u20101<\/jats:italic>)\u2010encoded MHC\u2010II transactivator (CIITA). This event is specific, as MHC class\u2004I remains unaffected. Similar results are observed with U937 cells expressing an exogenous full\u2010length CIITA. Molecular studies demonstrate that TPA treatment affects the stability of CIITA mRNA rather than CIITA transcription. Importantly, <jats:italic>cis<\/jats:italic>\u2010acting elements within the distal 650\u2004bp of the 1035\u2010bp 3\u2032\u2004untranslated region (3\u2032UTR, nucleotides 3509\u20134543) are associated to transcript instability. Transcription inhibitors actinomycin\u2004D and 5,6\u2010dichlororibofuranosyl benzimidazole, and the translation inhibitor cycloheximide significantly rescue the accumulation of CIITA mRNA in TPA\u2010treated cells. A similar effect is also observed after treatment with staurosporine and the PKC\u2010specific inhibitor GF109203X. The instability of CIITA mRNA produced by TPA in U937 cells is not seen in B\u2004cells.
