Previously, we found that <jats:italic>Salmonella choleraesuis<\/jats:italic> in combination with cisplatin could retard tumor growth in the murine subsutaneous hepatocellular carcinoma (HCC) model. In this regard, we investigated the antitumor activity of <jats:italic>S. choleraesuis<\/jats:italic> in the ML\u20101 orthotopic tumor model. Systemically administered <jats:italic>S. choleraesuis<\/jats:italic> accumulated within not only subcutaneous but also orthotopic tumors for at least 30 days, forming tumor\u2010to\u2010normal tissue ratios exceeding 1,000\u201310,000 to 1. The antitumor effects of <jats:italic>S. choleraesuis<\/jats:italic> were evaluated in mice bearing subcutaneous and orthotopic ML\u20101 tumors. Compared with the control treatment, <jats:italic>S. choleraesuis<\/jats:italic> significantly prolonged the animal survival, reduced the tumor size, as well as upregulated interferon (IFN)\u2010\u03b3 and induced IFN\u2010inducible chemokines CXCL10 (IP\u201010) productions. Furthermore, immunohistochemical staining of the tumors revealed decreased intratumoral microvessel density, increased infiltration of neutrophils, CD4<jats:sup>+<\/jats:sup> and CD8<jats:sup>+<\/jats:sup> T cells, and induced cell death in tumor microenvironment.
