{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2026,2,10]],"date-time":"2026-02-10T11:01:13Z","timestamp":1770721273191,"version":"3.49.0"},"reference-count":24,"publisher":"Wiley","issue":"2","license":[{"start":{"date-parts":[[2004,1,29]],"date-time":"2004-01-29T00:00:00Z","timestamp":1075334400000},"content-version":"vor","delay-in-days":5447,"URL":"http:\/\/onlinelibrary.wiley.com\/termsAndConditions#vor"}],"content-domain":{"domain":[],"crossmark-restriction":false},"short-container-title":["Yeast"],"published-print":{"date-parts":[[1989,3]]},"abstract":"<jats:title>Abstract<\/jats:title><jats:p>The secreted glycoproteins of <jats:italic>Pichia pastoris<\/jats:italic> contain more than 35% of their <jats:italic>N<\/jats:italic>\u2010linked oligosaccharides as structures smaller than Man<jats:sub>14<\/jats:sub>GlcNAc<jats:sub>2<\/jats:sub> (Man = mannose; GlcNAc = <jats:italic>N<\/jats:italic>\u2010acetylglucosamine). On heterologous invertase produced in <jats:italic>P. pastoris<\/jats:italic>, approximately 85% of the oligosaccharides are in the size range Man<jats:sub>8\u201314<\/jats:sub>GlcNAc<jats:sub>2<\/jats:sub>. The structures appear to contain \u03b1\u2010linked mannose. In addition, one\u2010third of the structures contain net negative charge and can be radio\u2010labelled <jats:italic>in vivo<\/jats:italic> with <jats:sup>32<\/jats:sup>P. The largest oligosaccharides isolated from <jats:italic>P. pastoris<\/jats:italic> are significantly shorter than the hypermannosylated structures typical of <jats:italic>S. cerevisiae<\/jats:italic>, indicating that the factors which influence the processing of <jats:italic>N<\/jats:italic>\u2010linked oligosaccharides in <jats:italic>P. pastoris<\/jats:italic> are different from those which influence processing in <jats:italic>S. cerevisiae.<\/jats:italic> The smaller <jats:italic>N<\/jats:italic>\u2010linked oligosaccharides synthesized by <jats:italic>P. pastoris<\/jats:italic> resemble high\u2010mannose oligosaccharides synthesized by animal cells, and this finding increases the utility of <jats:italic>P. pastoris<\/jats:italic> as a host for the production of heterologous glycoproteins.<\/jats:p>","DOI":"10.1002\/yea.320050206","type":"journal-article","created":{"date-parts":[[2005,5,28]],"date-time":"2005-05-28T23:24:20Z","timestamp":1117322660000},"page":"107-115","source":"Crossref","is-referenced-by-count":200,"title":["Size distribution and general structural features of <i>N<\/i>\u2010linked oligosaccharides from the methylotrophic yeast, <i>Pichia pastoris<\/i>"],"prefix":"10.1002","volume":"5","author":[{"given":"Lynn S.","family":"Grinna","sequence":"first","affiliation":[]},{"given":"Juerg F.","family":"Tschopp","sequence":"additional","affiliation":[]}],"member":"311","published-online":{"date-parts":[[2004,1,29]]},"reference":[{"key":"e_1_2_1_2_1","first-page":"335","volume-title":"The Molecular Biology of the Yeast Saccharomyces: Metabolism and Gene Expression","author":"Ballou C.
