Nevertheless, very active compounds have been identified in iris rhizomes including iridals and isoflavones.<\/jats:p><\/jats:sec><jats:sec><jats:title>Objective<\/jats:title><jats:p>In this first study concerning iris resinoid composition, flavonoids were qualitatively and quantitatively investigated in <jats:italic>I. germanica<\/jats:italic> and <jats:italic>I. pallida<\/jats:italic> resinoids.<\/jats:p><\/jats:sec><jats:sec><jats:title>Methodology<\/jats:title><jats:p>Resinoids were first fractionated by reverse\u2010phase flash chromatography in order to obtain fractions containing all isoflavones. These fractions were analysed by HPLC\u2010DAD (diode array detector) and the fractions containing isoflavones were analysed by HPLC\u2010QTOF (quadrupole time of flight)\u2010MS. Then, the main isoflavones were isolated and identified by NMR and high\u2010resolution mass spectroscopy (HRMS). Finally, total and individual isoflavones were quantified by HPLC\u2010DAD at 265\u2009nm using an external calibration method with irigenin as the external standard.<\/jats:p><\/jats:sec><jats:sec><jats:title>Results<\/jats:title><jats:p>Eight isoflavones were identified in both resinoids (irigenin, iristectorigenin A, nigricin, nigricanin, irisflorentin, iriskumaonin methyl ether, irilone, iriflogenin), one isoflavone only was identified in <jats:italic>I<\/jats:italic>. <jats:italic>germanica<\/jats:italic> resinoid (irisolidone), whereas one isoflavone (8\u2010hydroxyirigenin), one isoflavanone (2,3\u2010dihydroirigenin) and one benzophenone (2,6,4\u2032\u2010trihydroxy\u20104\u2010methoxybenzophenone) only were identified in <jats:italic>I<\/jats:italic>. <jats:italic>pallida<\/jats:italic> resinoid. Isoflavones were quantified in <jats:italic>I<\/jats:italic>. <jats:italic>germanica<\/jats:italic> and <jats:italic>I<\/jats:italic>. <jats:italic>pallida<\/jats:italic> resinoids at 180\u2009\u00b1\u20091.6\u2009mg\/g and 120\u2009\u00b1\u20093.3\u2009mg\/g respectively.<\/jats:p><\/jats:sec><jats:sec><jats:title>Conclusion<\/jats:title><jats:p>The study shows that <jats:italic>I<\/jats:italic>. <jats:italic>germanica<\/jats:italic> and <jats:italic>I<\/jats:italic>. <jats:italic>pallida<\/jats:italic> resinoids are rich in flavonoids and that these two <jats:italic>Iris<\/jats:italic> species can be distinguished by simply analysing the polyphenol fraction.
