However it was not possible to show this increased DNA synthesis by scintillometric measurement of the amount of <jats:sup>3<\/jats:sup>H\u2010TdR incorporated into extracted DNA. On the other hand, calcium did raise the incorporation of <jats:sup>14<\/jats:sup>C\u2010formate into the thymine residues of DNA, and increased the activity of isolated thymocyte thymidylate synthetase. In contrast to the mitogenic calcium ion, a thymidylate synthetase inhibitor, methotrexate, actually increased the incorporation of <jats:sup>3<\/jats:sup>H\u2010TdR into DNA.<\/jats:p><jats:p>It is concluded that calcium increases the endogenous synthesis of thymidylate which in turn prevents the amount of incorporation of exogenous <jats:sup>3<\/jats:sup>H\u2010TdR from accurately reflecting the true level of DNA synthesis.<\/jats:p>","DOI":"10.1002\/jcp.1040860306","type":"journal-article","created":{"date-parts":[[2005,2,26]],"date-time":"2005-02-26T17:39:34Z","timestamp":1109439574000},"page":"495-502","source":"Crossref","is-referenced-by-count":24,"title":["Failure of tritiated thymidine incorporation into DNA to reflect the autoradiographically demonstrable calcium\u2010induced increase in thymic lymphoblast DNA synthesis"],"prefix":"10.1002","volume":"86","author":[{"given":"T.","family":"Youdale","sequence":"first","affiliation":[]},{"given":"J.
