Herein, a microfluidic synthesis of self\u2010assembled carrier\u2010free gossypol (Gos)\u2010iron ion (Fe<jats:sup>3+<\/jats:sup>)\u2010toyocamycin (Toy) nanocapsules (GFT NCs) is developed for tumor magnetic resonance (MR) imaging and chemo\u2010chemodynamic\u2010immune therapy. The constructed GFT NCs with an average diameter of 101.9\u00a0nm display desirable colloidal stability, uniformity, pH\u2010responsive drug release profile, and enhanced <jats:italic>r<jats:sub>1<\/jats:sub><\/jats:italic> relaxivity (3.71\u00a0m<jats:sc>m<\/jats:sc><jats:sup>\u22121<\/jats:sup>s<jats:sup>\u22121<\/jats:sup>) under an acidic condition. The NCs\u2010mediated co\u2010delivery of Gos, Fe<jats:sup>3+<\/jats:sup>, and Toy induces apparent immunogenic cell death by leveraging a combination of Gos\u2010induced mitochondrial dysfunction, Fe<jats:sup>3+<\/jats:sup>\u2010mediated chemodynamic therapy, and Toy\u2010amplified endoplasmic reticulum stress, thus efficiently inhibiting tumor growth and metastasis while triggering a potent antitumor immunity. When combined with programmed cell death ligand 1 antibody, the GFT NCs significantly enhance immune activation and establish tumor\u2010specific immune memory effects, thereby preventing tumor recurrence. Compared to Magnevist, a commercial <jats:italic>T<\/jats:italic><jats:sub>1<\/jats:sub> MR imaging contrast agent, the GFT NCs offer superior tumor <jats:italic>T<\/jats:italic><jats:sub>1<\/jats:sub>\u2010weighted MR imaging at equivalent Fe or Gd concentrations in vivo. The developed GFT NCs represent a potential carrier\u2010free full\u2010active nanomedicine formulation for multifaceted and enhanced immunotherapy across diverse tumor
