A series of novel 3\u2010chloro\u20104\u2010[4\u2010(2\u2010oxo\u20102<jats:italic>H<\/jats:italic>\u2010chromen\u20104\u2010ylmethoxy)phenyl]\u20101\u2010phenylazetidin\u20102\u2010ones <jats:bold>5a\u2013o <\/jats:bold>have been synthesized from 4\u2010bromomethylcoumarins<jats:bold> 1a\u2013e<\/jats:bold> and 4\u2010aryliminomethyl\u2010phenols <jats:bold>3a\u2013c<\/jats:bold>. These compounds were screened for their <jats:italic>in\u2010vitro<\/jats:italic> antibacterial activity against two Gram\u2010positive (<jats:italic>Staphylococcus aureus<\/jats:italic> and<jats:italic> Vancomycin resistant enteroccoccus<\/jats:italic>) and two Gram\u2010negative (<jats:italic>Escherichia coli <\/jats:italic>and<jats:italic> Shigella dysentery<\/jats:italic>) bacterial strains and antifungal activity against<jats:italic> Aspergillus fumigatus<\/jats:italic>, <jats:italic>Candida albicans<\/jats:italic>, and<jats:italic> Penicillium.<\/jats:italic> Results revealed that compounds<jats:bold> 5c<\/jats:bold>,<jats:bold> 5f<\/jats:bold>, <jats:bold>5h<\/jats:bold>,<jats:bold> 5j<\/jats:bold>, and <jats:bold>5m <\/jats:bold>showed excellent activity against a panel of microorganisms. The brine\u2010shrimp bioassay was also carried out to study their<jats:italic> in\u2010vitro <\/jats:italic>cytotoxic properties and two compounds,<jats:bold> 5h <\/jats:bold>and<jats:bold> 5m<\/jats:bold>, possessing LD<jats:sub>50<\/jats:sub> = 7.154\u00d710<jats:sup>\u20134 <\/jats:sup>M and 5.782\u00d710<jats:sup>\u20134<\/jats:sup> M, respectively, displayed potent cytotoxic activity against <jats:italic>Artemia salina<\/jats:italic>. The presence of a chlorine group in the coumarin moiety, its effect on their antibacterial, antifungal, and cytotoxic activities is discussed.
