A detailed potentiometric, mass spectrometric and spectroscopic study showed that at a 1:1 ratio of C<jats:sub>Cu<\/jats:sub>\/C<jats:sub>peptide<\/jats:sub> this peptide formed a major [CuH(O<jats:sub>dPro<\/jats:sub>\uf8ffAsp)]<jats:sup>2+<\/jats:sup> species (pH range 5.5\u20137.0), in which the Cu<jats:sup>2+<\/jats:sup> ion was bound to the His and Asp residues in square\u2010planar or square\u2010pyramidal geometries. The stability constant corrected for protonated species (log\u2009<jats:italic>K*<\/jats:italic><jats:inline-graphic xmlns:xlink=\"http:\/\/www.w3.org\/1999\/xlink\" xlink:href=\"graphic\/tex2gif-inf-4.gif\" xlink:title=\"urn:x-wiley:09476539:media:CHEM201501715:tex2gif-inf-4\"\/>=9.33) is almost equal to the value obtained for the parent [CuH(O\uf8ffAsp)]<jats:sup>2+<\/jats:sup> species (log\u2009<jats:italic>K*<\/jats:italic><jats:sub>CuH(O\u2010Asp)<\/jats:sub>=9.28), but lower than that obtained for the cyclic [CuH(C\uf8ffAsp)]<jats:sup>2+<\/jats:sup> complex (log\u2009<jats:italic>K*<\/jats:italic><jats:sub>CuH(C\u2010Asp)<\/jats:sub>=10.79) previously published. Thus, the replacement of the ProGly unit by the stronger \u03b2\u2010turn\u2010inducing dProPro unit did not generate a more stable copper(II) species, although the O<jats:sub>dPro<\/jats:sub>\uf8ffAsp peptide was structured in solution, as shown by circular dichroism (CD) spectroscopy. Interestingly, the calculated value of <jats:italic>K<\/jats:italic><jats:sub>eff<\/jats:sub> showed that this peptide behaved similarly to the O\uf8ffAsp or C\uf8ffAsp counterparts, depending on the pH value. The cyclic voltammetry data indicated that the most easily reducible species were [CuH(O\uf8ffAsp)]<jats:sup>2+<\/jats:sup> (<jats:italic>E<\/jats:italic>\u2032<jats:sup>0<\/jats:sup>=262\u2005mV versus a normal hydrogen electrode (NHE)) and [CuH(O<jats:sub>dPro<\/jats:sub>\uf8ffAsp)]<jats:sup>2+<\/jats:sup> (<jats:italic>E<\/jats:italic>\u2032<jats:sup>0<\/jats:sup>=294\u2005mV versus NHE) complexes, the peptidic scaffolds of which are open. A lower value was obtained for [CuH(C\uf8ffAsp)]<jats:sup>2+<\/jats:sup> (<jats:italic>E<\/jats:italic>\u2032<jats:sup>0<\/jats:sup>=24\u2005mV versus NHE).
