Thirteen synthesized compounds were evaluated by radioligand binding (A<jats:sub>1<\/jats:sub>, A<jats:sub>2A<\/jats:sub>, and A<jats:sub>3<\/jats:sub>) and adenylyl cyclase activity (A<jats:sub>2B<\/jats:sub>) assays in order to study their affinity for the four human AR (hAR) subtypes. Seven of the studied compounds proved to be selective A<jats:sub>3<\/jats:sub>AR ligands, with 3\u2010(4\u2032\u2010methylphenyl)\u20108\u2010(2\u2010oxopropoxy)coumarin (<jats:bold>12<\/jats:bold>) being the most potent (<jats:italic>K<\/jats:italic><jats:sub>i<\/jats:sub>=634\u2005n<jats:sc>M<\/jats:sc>). None of the compounds showed affinity for the A<jats:sub>2B<\/jats:sub> receptor, while four compounds were found to be nonselective AR ligands for the other three subtypes. Docking simulations were carried out to identify the hypothetical binding mode and to rationalize the interaction of these types of coumarin derivatives with the binding site of the three ARs to which binding was observed. The results allowed us to conclude that the 3\u2010arylcoumarin scaffold composes a novel and promising class of A<jats:sub>3<\/jats:sub>AR ligands.
