Novel hydroxylated 3\u2010arylcoumarins were designed, synthesized, and evaluated for their XO inhibition and antioxidant properties. 3\u2010(3\u2019\u2010Bromophenyl)\u20105,7\u2010dihydroxycoumarin (compound <jats:bold>11<\/jats:bold>) proved to be the most potent XO inhibitor, with an IC<jats:sub>50<\/jats:sub> of 91\u2005nM, being 162 times better than allopurinol, one of the reference controls. Kinetic analysis of compound <jats:bold>11<\/jats:bold> and compound <jats:bold>5<\/jats:bold> [3\u2010(4\u2019\u2010bromothien\u20102\u2019\u2010yl)\u20105,7\u2010dihydroxycoumarin], the second\u2010best compound within the series (IC<jats:sub>50<\/jats:sub> of 280\u2005nM), has been performed, and both compounds showed a mixed\u2010type inhibition. Both compounds present good antioxidant activity (ability to scavenge ABTS radical) and are able to reduce reactive oxygen species (ROS) levels in H<jats:sub>2<\/jats:sub>O<jats:sub>2<\/jats:sub>\u2010treated cells. In addition, they proved to be non\u2010cytotoxic in a Caco\u20102 cells viability assay.
