{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2026,1,28]],"date-time":"2026-01-28T21:47:17Z","timestamp":1769636837140,"version":"3.49.0"},"reference-count":46,"publisher":"Wiley","issue":"24","license":[{"start":{"date-parts":[[2024,11,5]],"date-time":"2024-11-05T00:00:00Z","timestamp":1730764800000},"content-version":"vor","delay-in-days":0,"URL":"http:\/\/onlinelibrary.wiley.com\/termsAndConditions#vor"}],"content-domain":{"domain":["chemistry-europe.onlinelibrary.wiley.com"],"crossmark-restriction":true},"short-container-title":["ChemMedChem"],"published-print":{"date-parts":[[2024,12,16]]},"abstract":"<jats:title>Abstract<\/jats:title><jats:p>We herein describe the radiosynthesis of a <jats:sup>125<\/jats:sup>I\u2010labeled acridine orange derivative (<jats:bold>[<jats:sup>125<\/jats:sup>I]\u2010C<jats:sub>8<\/jats:sub><\/jats:bold>), acting as a G\u2010quadruplex binder, and its biological evaluation in cervical cancer models, aiming to enlighten its potential as a radioligand for Auger Electron Radiopharmaceutical Therapy (AE\u2010RPT) of cancer. <jats:bold>[<jats:sup>125<\/jats:sup>I]\u2010C<jats:sub>8<\/jats:sub><\/jats:bold> was synthesized with a moderate radiochemical yield (ca. 60\u2009%) by a [<jats:sup>125<\/jats:sup>I]iodo\u2010destannylation reaction. Its evaluation in cervical cancer HeLa cells demonstrated that the radiocompound has a significant cellular internalization with a notorious accumulation in the cell nucleus. In line with these results, <jats:bold>[<jats:sup>125<\/jats:sup>I]\u2010C<jats:sub>8<\/jats:sub><\/jats:bold> strongly compromised the viability of HeLa cells in a dose\u2010dependent manner, inducing non\u2010repairable DNA lesions that are most probably due to the AEs emitted by <jats:sup>125<\/jats:sup>I in close proximity to the DNA molecule. Biodistribution studies in a murine HeLa xenograft model showed that <jats:bold>[<jats:sup>125<\/jats:sup>I]\u2010C<jats:sub>8<\/jats:sub><\/jats:bold> has fast blood clearance and high <jats:italic>in\u2005vivo<\/jats:italic> stability but poor tumor uptake, after systemic administration. The respective supramolecular conjugate with the AS1411 aptamer (<jats:bold>[<jats:sup>125<\/jats:sup>I]\u2010C<jats:sub>8<\/jats:sub>\/AS1411<\/jats:bold>) led to a slower blood clearance in the same animal tumor model, although without improving the tumor uptake. To take advantage of the radiotoxicity of <jats:bold>[<jats:sup>125<\/jats:sup>I]\u2010C<jats:sub>8<\/jats:sub><\/jats:bold> against cervical cancer cells other strategies need to be studied, based namely on alternative nanodelivery carriers and\/or intratumoral injection
