In order to promote regeneration, the process should result in a peripheral T\u2010cell pool with a similar subpopulation structure as before lymphopenia. We investigated the repopulation of the CD8<jats:sup>+<\/jats:sup> central\u2010memory T cells (T<jats:sub>CM<\/jats:sub>) and effector\u2010memory T cells (T<jats:sub>EM<\/jats:sub>) pools after adoptive transfer of sorted CD8<jats:sup>+<\/jats:sup> T cells from na\u00efve, T<jats:sub>CM<\/jats:sub> and T<jats:sub>EM<\/jats:sub> subsets into T\u2010cell\u2010deficient hosts. We show that the initial kinetics of expansion are distinct for each subset and that the contribution to the repopulation of the CD8<jats:sup>+<\/jats:sup> T\u2010cell pool by the progeny of each subset is not a mere function of its initial expansion. We demonstrate that CD4<jats:sup>+<\/jats:sup>CD25<jats:sup>+<\/jats:sup> Treg play a major role in the repopulation of the CD8<jats:sup>+<\/jats:sup> T\u2010cell pool and that CD8<jats:sup>+<\/jats:sup> T\u2010cell subsets impact on each other. In the absence of CD4<jats:sup>+<\/jats:sup>CD25<jats:sup>+<\/jats:sup> Treg, a small fraction of na\u00efve CD8<jats:sup>+<\/jats:sup> T cells strongly proliferates, correlating with further expansion and differentiation of co\u2010expanding CD8<jats:sup>+<\/jats:sup> T cells.
