CLEC\u20102 belongs to the Dectin\u20101 sub\u2010family of CLRs that possess an extracellular C\u2010type lectin\u2010like domain and a single intracellular hemITAM motif. CLEC\u20102 is highly expressed on mouse and human platelets where it signals via Syk to promote aggregation. We generated a monoclonal antibody (mAb) against mouse CLEC\u20102 and found that CLEC\u20102 is additionally widely expressed on leukocytes and that its expression is upregulated during inflammation. MAb\u2010mediated crosslinking of CLEC\u20102 leads to hemITAM\u2010dependent signaling via Syk, Ca<jats:sup>2+<\/jats:sup> and NFAT and, in myeloid cells, modulates the effect of toll\u2010like receptor (TLR) agonists to selectively potentiate production of IL\u201010. A macrophage\/dendritic cell\u2010dependent increase in IL\u201010 is also observed in mice given anti\u2010CLEC\u20102 mAb together with LPS. Collectively, these data indicate that CLEC\u20102 is expressed in myeloid cells and acts as a Syk\u2010coupled CLR able to modulate TLR signaling and inflammatory
