For further standardization of this method, we analyzed cerebrospinal fluid (CSF) of eight probable AD patients and seven nondemented controls using different preanalytical procedures for A\u03b2\u2010SDS\u2010PAGE\/immunoblot and A\u03b21\u201042\u2010enzyme linked immunosorbent assay (ELISA). Both diagnostic groups were discriminated significantly by absolute levels of A\u03b21\u201042 and ratios of A\u03b21\u201042\/40, 1\u201042\/38, 1\u201042\/39. Preanalytical freezing of CSF led to a highly significant loss of all A\u03b2 peptide species. This effect was most pronounced for\u00a0A\u03b21\u201042 and completely prevented by SDS\u2010heat denaturation prior to freezing. Prolonged storage of SDS\u2010heat denatured CSF led to a selective loss of A\u03b21\u201042 and impaired the discrimination of diagnostic groups as measured by A\u03b2\u2010SDS\u2010PAGE\/immunoblot. Neither freezing nor storage significantly affected absolute A\u03b21\u201042 levels as determined by A\u03b21\u201042\u2010ELISA, but both impaired the discrimation of diagnostic groups. Hence, we suggest immediate analysis of samples for A\u03b21\u201042\u2010ELISA, analysis after a short freezing interval for A\u03b2\u2010SDS\u2010PAGE\/immunoblot, and avoidance of prolonged storage intervals. Remarkably, A\u03b2\u2010SDS\u2010PAGE\/immunoblot measured threefold higher levels of A\u03b21\u201042 in CSF than A\u03b21\u201042\u2010ELISA.
