In the past, <jats:italic><jats:styled-content style=\"fixed-case\">T<\/jats:styled-content>heileria<\/jats:italic> infection in cattle in <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>ustralia was largely asymptomatic and recognized to be associated with <jats:italic><jats:styled-content style=\"fixed-case\">T<\/jats:styled-content>heileria buffeli<\/jats:italic>. However, outbreaks of theileriosis have occurred in beef and dairy cattle in subtropical climatic regions (<jats:styled-content style=\"fixed-case\">N<\/jats:styled-content>ew <jats:styled-content style=\"fixed-case\">S<\/jats:styled-content>outh <jats:styled-content style=\"fixed-case\">W<\/jats:styled-content>ales) of <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>ustralia. There is also one published report of a recent theileriosis outbreak in a beef farm near <jats:styled-content style=\"fixed-case\">S<\/jats:styled-content>eymour in the southeastern state of <jats:styled-content style=\"fixed-case\">V<\/jats:styled-content>ictoria. In order to gain an improved insight into the genetic composition of <jats:italic><jats:styled-content style=\"fixed-case\">T<\/jats:styled-content>heileria<\/jats:italic> populations following this outbreak, we undertook herein an integrated <jats:styled-content style=\"fixed-case\">PCR<\/jats:styled-content>\u2010coupled mutation scanning\u2010sequencing\u2010phylogenetic analysis of sequence variation in part of the major piroplasm surface protein (<jats:styled-content style=\"fixed-case\">MPSP<\/jats:styled-content>) gene within and among samples from cattle involved in the outbreak. <jats:italic>Theileria<\/jats:italic> <jats:styled-content style=\"fixed-case\">DNA<\/jats:styled-content> was detected in 89.4% of 94 cattle in the <jats:styled-content style=\"fixed-case\">S<\/jats:styled-content>eymour farm; the genetic analysis showed that the <jats:italic>ikeda<\/jats:italic> and <jats:italic>chitose<\/jats:italic> genotypes representing the <jats:italic><jats:styled-content style=\"fixed-case\">T<\/jats:styled-content>heileria orientalis<\/jats:italic> complex were detected in 75 and 4.8% of 84 infected cattle, respectively, and that mixed populations of these two genotypes were found in 20.2% of infected cattle. Given unpublished reports of a significant increase in the number of outbreaks in <jats:styled-content style=\"fixed-case\">V<\/jats:styled-content>ictoria, future investigations should focus sharply on elucidating the epidemiology of <jats:italic><jats:styled-content style=\"fixed-case\">T<\/jats:styled-content>heileria<\/jats:italic> to subvert the economic impact on the cattle industry in this state.
