Basic Microbiol."],"published-print":{"date-parts":[[2015,10]]},"abstract":"<jats:sec><jats:label\/><jats:p>This study aimed at the isolation and characterization of metal(loid)\u2010tolerant bacteria from the rhizosphere of <jats:italic>Phragmites australis<\/jats:italic> and <jats:italic>Juncus effusus<\/jats:italic> plants growing in two long\u2010term contaminated sites in Northern Portugal. Site 1 had higher contamination than Site 3. Bacteria were isolated using metal(loid)\u2010supplemented (Cd, Zn, and As) media. Isolates were grouped by random amplified polymorphic DNA and identified by 16S rRNA gene sequencing. Strains were also examined for their metal(loid) tolerance. The counts of metal(loid)\u2010tolerant bacteria were higher in Site 1 and ranged between log 7.17\u2009CFU\u2009g<jats:sup>\u22121<\/jats:sup> soil in As\u2010containing medium and log 7.57\u2009CFU\u2009g<jats:sup>\u22121<\/jats:sup> soil in Zn\u2010containing medium, while counts at Site 3 varied between log 5.33\u2009CFU\u2009g<jats:sup>\u22121<\/jats:sup> soil in Cd\u2010containing medium and log 6.97\u2009CFU\u2009g<jats:sup>\u22121<\/jats:sup> soil in As\u2010containing medium. The composition of bacterial populations varied between locations. In Site 1, the classes <jats:italic>Actinobacteria<\/jats:italic> (36%) and <jats:italic>Bacilli<\/jats:italic> (24%) were well represented, while in Site 3 strains were mainly affiliated to classes <jats:italic>Actinobacteria<\/jats:italic> (35%), \u03b3\u2010<jats:italic>Proteobacteria<\/jats:italic> (35%), and \u03b2\u2010<jats:italic>Proteobacteria<\/jats:italic> (12%). The order of metal(loid) toxicity for the isolated strains was Cd\u2009&gt;\u2009As\u2009&gt;\u2009Zn. Overall, 10 strains grew at 500\u2009mg\u2009Cd\u2009L<jats:sup>\u22121<\/jats:sup>, 1000\u2009mg\u2009Zn\u2009L<jats:sup>\u22121<\/jats:sup>, and 500\u2009mg\u2009As\u2009L<jats:sup>\u22121<\/jats:sup>, being considered the most metal(loid)\u2010tolerant bacteria. These strains belonged to genera <jats:italic>Cupriavidus<\/jats:italic>, <jats:italic>Burkholderia<\/jats:italic>, <jats:italic>Novosphingobium<\/jats:italic>, <jats:italic>Sphingobacterium<\/jats:italic>, <jats:italic>Castellaniella<\/jats:italic>, <jats:italic>Mesorhizobium<\/jats:italic>, <jats:italic>Chryseobacterium<\/jats:italic>, and <jats:italic>Rhodococcus<\/jats:italic> and were mainly retrieved from Site 1.
