The enzyme contains two <jats:italic>c<\/jats:italic>\u2010type haems: one is reducible physiologically by cytochrome <jats:italic>c<\/jats:italic><jats:sub>550<\/jats:sub> from the same organism or non\u2010physiologically by ascorbate (high\u2010potential haem) and the other by dithionite (low\u2010potential haem). The enzymatically active form of the peroxidase is the half\u2010reduced enzyme state, in which the high\u2010potential haem is in the iron(II) state and the low\u2010potential haem is in the iron(III) state. It was found that the two haems interact and that the enzyme binds calcium ions near the haem sites which are necessary to promote its activation. In the oxidized form, the high\u2010potential haem is in a high\u2010spin and the low\u2010potential haem is in a low\u2010spin state. The half\u2010reduction of the enzyme with ascorbate\u2010diaminodurol changes the high\u2010potential haem (high\u2010spin) into a low\u2010spin state and the low\u2010potential haem converts from a low\u2010 into a high\u2010spin state. This high\u2010spin conversion of the low\u2010potential haem is induced by the presence of calcium ions.
