By combining high\u2010yield A\u03b2 immuno\u00ad precipitation (IP), IEF, and urea\u2010based A\u03b2\u2010SDS\u2010PAGE\u2010immunoblot, at least 30 A\u03b2\u2010immuno\u00adreactive spots were detected in human plasma samples as small as 1.6\u2005mL. This approach clearly resolved A\u03b2 peptides A\u03b2<jats:sub>1\u201340<\/jats:sub>, A\u03b2<jats:sub>1\u201042<\/jats:sub>, A\u03b2<jats:sub>1\u201037<\/jats:sub>, A\u03b2<jats:sub>1\u201038<\/jats:sub>, A\u03b2<jats:sub>1\u201039<\/jats:sub>, the N\u2010truncated A\u03b2<jats:sub>2\u201340<\/jats:sub>, A\u03b2<jats:sub>2\u201342<\/jats:sub>, and, for the first time, also A\u03b2<jats:sub>1\u201341<\/jats:sub>. Relative quantification indicated that A\u03b2<jats:sub>1\u201340<\/jats:sub> and A\u03b2<jats:sub>1\u201342<\/jats:sub> accounted for less than 60% of the total amount of A\u03b2 peptides in plasma. All other A\u03b2 peptides appear to be either C\u2010terminally or N\u2010terminally truncated forms or as yet uncharacterized A\u03b2 species which migrated as trains of spots with distinct p<jats:italic>I<\/jats:italic>s. The A\u03b2 pattern found in cerebrospinal fluid (CSF) was substantially less complex. This sensitive method (2\u2010D A\u03b2\u2010WIB) might help clarifying the origin of distinct A\u03b2 species from different tissues, cell types, or intracellular pools as well as their amyloidogenicity.
