The objective of the present study was to characterize aminoterminally truncated or oxidized <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b238, <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b240, and <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b242 peptide species in immunoprecipitated human cerebrospinal fluid (<jats:styled-content style=\"fixed-case\">CSF<\/jats:styled-content>).<\/jats:p><\/jats:sec><jats:sec><jats:title>Experimental design<\/jats:title><jats:p>We invented a novel sequential aminoterminally and carboxyterminally specific immunoprecipitation protocol and used the <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b2\u2010SDS\u2010PAGE\/immunoblot for subsequent analysis of <jats:styled-content style=\"fixed-case\">CSF<\/jats:styled-content> <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b2 peptide patterns.<\/jats:p><\/jats:sec><jats:sec><jats:title>Results<\/jats:title><jats:p>In the present study, we identified the aminoterminally truncated <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b2 peptides 2\u201340 and 2\u201342 as well as oxidized forms of <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b21\u201338 and <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b21\u201342 in <jats:styled-content style=\"fixed-case\">CSF<\/jats:styled-content>. Our protocol allowed the quantification of a pattern of <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b2 peptides 1\u201338<jats:sup>ox<\/jats:sup>, 2\u201340, and 2\u201342 in addition to the well known panel of <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b2 1\u201337, 1\u201338, 1\u201339, 1\u201340, 1\u201340<jats:sup>ox<\/jats:sup>, and 1\u201342 in a group of seven patients with peripheral polyneuropathy.<\/jats:p><\/jats:sec><jats:sec><jats:title>Conclusions and clinical relevance<\/jats:title><jats:p>In the present approach, we could broaden the range of quantifiable <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b2 peptides described in previous studies (i.e., 1\u201337, 1\u201338, 1\u201339, 1\u201340, 1\u201340<jats:sup>ox<\/jats:sup>, and 1\u201342) by <jats:styled-content style=\"fixed-case\">A<\/jats:styled-content>\u03b2 1\u201338<jats:sup>ox<\/jats:sup>, 2\u201340, and 2\u201342.
