It is based on the electrophoretic pattern of total tRNA samples (a \u2018tRNA profile\u2019) isolated from <jats:italic>Candida<\/jats:italic> species and generated using high\u2010resolution semi\u2010denaturing urea\u2010polyacrylamide gel electrophoresis and methylene blue staining. Species\u2010specific tRNA profiles for the species. <jats:italic>C. albicans<\/jats:italic>, <jats:italic>C. tropicalis<\/jats:italic>, <jats:italic>C. parapsilosis<\/jats:italic>, <jats:italic>C. guilliermondii<\/jats:italic>, <jats:italic>C. glabrata<\/jats:italic> and <jats:italic>Pichia guilliermondii<\/jats:italic> were obtained. Detailed studies with the major human pathogen of the <jats:italic>Candida<\/jats:italic> genus, <jats:italic>C. albicans<\/jats:italic>, demonstrated that the tRNA profile for a given species was both reproducible and strain\u2010independent; seven different <jats:italic>C. albicans<\/jats:italic> strains generated identical tRNA profiles. Minor strain\u2010specific heterogeneities in the tRNA profiles of <jats:italic>C. guilliermondii<\/jats:italic> and <jats:italic>C. parapsilosis<\/jats:italic> were detected, but in neither case did they significantly alter the species\u2010specific diagnostic tRNA profile. The potential of this method in clarifying taxonomic anomalies was demonstrated by the finding that Type I and Type II strains of <jats:italic>C. stellatoidea<\/jats:italic> generate very different tRNA profiles, with that of a Type II strain being identical to the <jats:italic>C. albicans<\/jats:italic> tRNA profile.
