This InsP<jats:sub>4<\/jats:sub> isomer accumulated in vasopressin stimulated hepatocytes prelabeled with <jats:italic>myo\u2010<\/jats:italic>[<jats:sup>3<\/jats:sup>H]inositol with a time course that lagged behind Ins 1,3,4\u2010P<jats:sub>3<\/jats:sub> formation. The Ins 1,3,4\u2010P<jats:sub>3<\/jats:sub> kinase responsible for its formation was partially purified from rat liver. The enzyme had a <jats:italic>K<\/jats:italic>\n<jats:sub>m<\/jats:sub> for Ins 1,3,4\u2010P<jats:sub>3<\/jats:sub> of 0.29 \u03bcM, a <jats:italic>K<\/jats:italic>\n<jats:sub>m<\/jats:sub> for ATP of 141 \u03bcM and was not affected by changes in free Ca<jats:sup>2+<\/jats:sup> in the physiological range.
