Rank order of binding activities was: GTP\u03b3S&gt;GTP&gt;8\u2010N<jats:sub>3<\/jats:sub>\u2010GTP; the binding of GTP\u03b3S and GTP, but not of 8\u2010N<jats:sub>3<\/jats:sub>\u2010GTP, was stimulated by receptor agonists. [<jats:sup>3<\/jats:sup>H]GTP binding to <jats:italic>D. discoideum<\/jats:italic> membranes has been described previously by a single binding type (<jats:italic>K<\/jats:italic>\n<jats:sub>d<\/jats:sub> = 2.6 \u03bcM, <jats:italic>B<\/jats:italic>\n<jats:sub>max<\/jats:sub> = 85 nM). More detailed studies with [<jats:sup>35<\/jats:sup>S]GTP\u03b3S showed heterogeneous binding composed of two forms of binding sites with respectively high (<jats:italic>K<\/jats:italic>\n<jats:sub>d<\/jats:sub> = 0.2 \u03bcM) and low (<jats:italic>K<\/jats:italic>\n<jats:sub>d<\/jats:sub> = 6.3 \u03bcM) affinity. cAMP derivatives enhanced GTP\u03b3S binding by increasing the affinity and the number of the high\u2010affinity sites, while the low\u2010affinity sites were not affected by cAMP. The specificity of cAMP derivatives for stimulation of GTP\u03b3S binding showed aclose correlation with the specificity for binding to the cell surface cAMP receptor. Pretreatment of <jats:italic>D. discoideum<\/jats:italic> cells with pertussis toxin did not affect basal GTP and GTP\u03b3S binding, but eliminated the cAMP stimulation of GTP and GTP\u03b3S binding.
