To identify the <jats:italic>CDC25<\/jats:italic> gene product, we raised antibodies against two hybrid proteins, encoded by in\u2010frame fusions between the <jats:italic>E. coli lacZ<\/jats:italic> gene and two different parts of the <jats:italic>CDC25<\/jats:italic> gene. By protein immuno\u2010blotting, we were able to identify the <jats:italic>CDC25<\/jats:italic> gene product as a 180 kDa polypeptide, which we named p180<jats:sup>CDC25<\/jats:sup>. It was detected only when the <jats:italic>CDC25<\/jats:italic> gene was overexpressed in a proteases\u2010deficient yeast strain. Subcellular fractionation experiments showed that p180<jats:sup>CDC25<\/jats:sup>, as well as ras proteins, is attached to the membrane, even after treatments which release peripheral membrane
