It resembles the catabolite gene activator or cAMP\u2010receptor protein (CRP) except for the presence of an N\u2010terminal cysteine cluster, which may form a redox\u2010sensing iron\u2010binding site. Site\u2010directed mutagenesis has shown that 3 of the 4 cysteine residues in the N\u2010terminal cluster (Cys\u201020, \u201023 and \u201029, but not Cys\u201016) and the only other cysteine residue (Cys\u2010122), are essential for the normal activation and repression of PNR\u2010dependent promoters. Deletion of residues Pro\u20103\u2010Arg\u20109 (inclusive) had no effect, but FNR was inactivated by a frameshift extending through the C\u2010terminal DNA\u2010binding domain.
