We found that micromolar concentration of a synthetic peptide, corresponding to residues 312\u2013326 of the PP\u20101 carboxyl terminus (P312\u2010326) that is missing from PP\u201035K, increased the phosphatase activity of PP\u201035K with phosphorylase and myosin light chains as substrates by decreasing the apparent <jats:italic>K<\/jats:italic>\n<jats:sub>m<\/jats:sub> without a change in <jats:italic>V<\/jats:italic>\n<jats:sub>m<\/jats:sub>. Purified PP\u20101 and PP\u201035K were inhibited identically by okadaic acid, but peptide P312\u2010326 only stimulated the activity of PP\u201035K, not full\u2010length PP\u20101. Other peptides corresponding to the carboxyl terminus of phosphatase\u20102A or to the amino terminus of PP\u20101 did not affect the activity of PP\u201035K. A sequence conserved in PP\u20101 from different species, Pro\u2010He\u2010Thr\u2010Pro\u2010Pro was implicated as the active region because a derivative peptide, Ala\u2010Pro\u2010Ile\u2010Thr\u2010Pro\u2010Pro\u2010Ala, stimulated the activity of PP\u201035K to the same extent as peptide P312\u2010326 although at higher concentrations.
