Site\u2010specific reaction of individual GTP\u2010binding proteins was achieved by cross\u2010linking the protein\u2010bound 2\u2032,3\u2032\u2010dialdehyde derivative of GTP with the single lysine residue of the conserved NKXD sequence through Schiff's base formation and subsequent cyanoborohydride reduction. Labeled GTP\u2010binding proteins from prokaryotic or eukaryotic cell homogenates were separated by polyacrylamide gel electrophoresis and visualized by autoradiography. In addition cross\u2010linking of [\u03b1\u2010<jats:sup>32<\/jats:sup>P]GTP with GTP\u2010binding proteins was demonstrated in model systems using different purified GTPases, human c\u2010H\u2010<jats:italic>ras<\/jats:italic> p21, transducin from bovine retina, polypeptide elongation factor Tu (EF\u2010Tu) from <jats:italic>\nT. themophilus<\/jats:italic> and initiation factor 2 (1F2) from <jats:italic>T. thermophilus<\/jats:italic>.
