In addition, unaltered Gly positions could have a crucial role in the building of the structural framework. We have modified <jats:italic>Drosophila<\/jats:italic> ADH and expressed the mutant forms in <jats:italic>E. coli<\/jats:italic>. Mutation of Tyr<jats:sup>152<\/jats:sup> to Glu or Gin, Lys<jats:sup>156<\/jats:sup> to Ile, Gly<jats:sup>184<\/jats:sup> to Leu, and the double mutant Gly<jats:sup>130<\/jats:sup> to Cys and Gly<jats:sup>133<\/jats:sup> to Ile, all rendered, with different substrates and at different pHs, an inactive enzyme.
