We expressed <jats:italic>AHA2<\/jats:italic>, a member of the <jats:italic>Arabidopsis thaliana<\/jats:italic> pm H<jats:sup>+<\/jats:sup>\u2010ATPase gene family, in yeast cells in which transcription of the endogenous pm H<jats:sup>+<\/jats:sup>ATPase gene (<jats:italic>pmal<\/jats:italic>) had been turned off. <jats:italic>AHA2<\/jats:italic> was expressed mainly in intracellular membranes and only supported very slow growth of transformed yeast cells. Removal of the last 92 C\u2010terminal amino acids from the plant H<jats:sup>+<\/jats:sup>\u2010ATPase produced an enzyme with 2\u20133\u2010fold higher specific ATPase activity than the wild\u2010type plant enzyme.
