From the N\u2010terminal amino acid sequences of each subunit and its polypeptide fragment obtained by partial digestion with V8 protease, oligonucleotides corresponding to forward and reverse primers for each gene (<jats:italic>NQR A<\/jats:italic>, <jats:italic>B<\/jats:italic> and <jats:italic>C<\/jats:italic>) encoding the \u03b1, \u03b2 and \u03b3 subunit, respectively, were synthesized. Using these primers, a part of each gene was amplified from the chromosomal DNA of <jats:italic>V. alginolyticus<\/jats:italic> by a PCR method, and the PCR products were used for the cloning of the <jats:italic>NQR<\/jats:italic> gene in \u03bb phage.
