A null mutation in <jats:italic>ace\u20101<\/jats:italic> (allele <jats:italic>p1000<\/jats:italic>) suppresses all acetylcholinesterase activity of class A. We have identified an opal mutation TG<jats:inline-graphic xmlns:xlink=\"http:\/\/www.w3.org\/1999\/xlink\" xlink:href=\"graphic\/feb2001457939401343y-math-si1.gif\" xlink:title=\"urn:x-wiley:00145793:media:feb2001457939401343y:feb2001457939401343y-math-si1\" \/> (W99)\u2192TG<jats:inline-graphic xmlns:xlink=\"http:\/\/www.w3.org\/1999\/xlink\" xlink:href=\"graphic\/feb2001457939401343y-math-si2.gif\" xlink:title=\"urn:x-wiley:00145793:media:feb2001457939401343y:feb2001457939401343y-math-si2\" \/> (Stop) as the only alteration in the mutated gene. This leads to a truncated protein (98 instead of 620 amino acids) with no enzymatic activity. The mutation also reduces the level of <jats:italic>ace\u20101<\/jats:italic> transcripts to only 10% of that in wild\u2010type animals. This most likely results from a destabilization of mRNA containing the nonsense message.
