SNAP\u201025 occurs in neuroendocrine cells and, in analogy to its role in neurons, has been implicated in catecholamine secretion, yet the nature of the underlying mechanism remains obscure. Here we use an anti\u2010SNAP\u201025 monoclonal antibody to show that SNAP\u201025 is localized at the cytosolic surface of the plasma membrane of chromaffin cells. This antibody inhibited the Ca<jats:sup>2+<\/jats:sup>\u2010evoked catecholamine release from digitonin\u2010permeabilized chromaffin cells in a time\u2010 and dose\u2010dependent manner. Remarkably, a 20\u2010mer synthetic peptide representing the sequence of the C\u2010terminal domain of SNAP\u201025 blocked Ca<jats:sup>2+<\/jats:sup>\u2010dependent catecholamine release with an IC<jats:sub>50<\/jats:sub> = 20 \u03bcM. The inhibitory activity of the peptide was sequence\u2010specific as evidenced by the inertness of a control peptide with the same amino acid composition but random order.
