The altered proteins were overproduced in <jats:italic>Escherichia coli<\/jats:italic>, purified and characterized. The EF\u2010Tu T62S variant had similar properties with respect to thermostability, aminoacyl\u2010tRNA binding, GTPase activity and in vitro translation as the wild\u2010type EF\u2010Tu. In contrast, EF\u2010Tu T62A is severely impaired in its ability to sustain polypeptide synthesis and has only very low intrinsic and ribosome\u2010induced GTPase activity. The affinity of aminoacyl\u2010tRNA to the EF\u2010Tu T62A\u00b7GTP complex is almost 40 times lower as compared to the native EF\u2010Tu\u00b7GTP. These observations are in agreement with the tertiary structure of EF\u2010Tu\u00b7GTP, in which threonine\u201062 is interacting with the Mg<jats:sup>2+<\/jats:sup> ion, \u03b3\u2010phosphate of GTP and a water molecule, which is presumably involved in the GTP
