Mutations in <jats:italic>OXA1<\/jats:italic> lead to a complete respiratory deficiency. The protein Oxa1p is conserved through evolution and a human homolog has been isolated by functional complementation of a yeast <jats:italic>oxa1<jats:sup>\u2212<\/jats:sup>\n<\/jats:italic> mutant. In order to further our understanding of the role of Oxa1p, we have constructed two yeast strains in which the <jats:italic>OXA1<\/jats:italic> open reading frame was almost totally deleted. Cytochrome spectra and enzymatic activity measurements show the absence of heme <jats:italic>aa<\/jats:italic>\n<jats:sub>3<\/jats:sub> and of a cytochrome <jats:italic>c<\/jats:italic> oxido\u2010reductase activity and dramatic decrease of the oligomycin sensitive ATPase activity.
