CBFs consist of a DNA binding CBF\u03b1 subunit and a non\u2010DNA binding CBF\u03b2 subunit that increases the affinity of CBF\u03b1 for DNA. We performed sedimentation equilibrium analyses to unequivocally establish the stoichiometry of the CBF\u03b1:\u03b2:DNA complex. Dissociation constants for all four equilibria involving the CBF\u03b1 Runt domain, CBF\u03b2, and DNA were defined. Conformational changes associated with interactions between CBF\u03b1, CBF\u03b2, and DNA were monitored by nuclear magnetic resonance and circular dichroism spectroscopy.
