We found that H<jats:sub>2<\/jats:sub>O<jats:sub>2<\/jats:sub> inhibited the activity of recombinant caspase\u20103 and caspase\u20108, half\u2010inhibition occurring at about 17 \u03bcM H<jats:sub>2<\/jats:sub>O<jats:sub>2<\/jats:sub>. This inhibition was both prevented and reversed by dithiothreitol while glutathione had little protective effect. 100\u2013200 \u03bcM H<jats:sub>2<\/jats:sub>O<jats:sub>2<\/jats:sub> added to macrophages after induction of caspase activation by nitric oxide or serum withdrawal substantially inhibited caspase activity. Activation of H<jats:sub>2<\/jats:sub>O<jats:sub>2<\/jats:sub>\u2010producing NADPH oxidase in macrophages also caused catalase\u2010sensitive inactivation of cellular caspases.
