We were interested in investigating whether an NO\u2010generating system could be present in plants. NO and <jats:sc>l<\/jats:sc>\u2010[<jats:sup>14<\/jats:sup>C]citrulline were synthesized by roots and nodules of <jats:italic>Lupinus albus<\/jats:italic> in an <jats:sc>l<\/jats:sc>\u2010arginine\u2010dependent manner. <jats:sc>l<\/jats:sc>\u2010[<jats:sup>14<\/jats:sup>C]Citrulline production was inhibited by <jats:italic>N<\/jats:italic><jats:sup>G<\/jats:sup>\u2010monomethyl\u2010<jats:sc>l<\/jats:sc>\u2010arginine, a nitric oxide synthase antagonist, in a competitive way. NADPH\u2010diaphorase activity was localized in the vascular bundles in root and nodules, and also in the nodule infected zone. This staining was significantly reduced in the presence of <jats:italic>N<\/jats:italic><jats:sup>G<\/jats:sup>\u2010monomethyl\u2010<jats:sc>l<\/jats:sc>\u2010arginine.
