The minimum sequence motif required for efficient phosphorylation was Arg\u2010Xaa\u2010Arg\u2010Yaa\u2010Zaa\u2010Ser\/Thr\u2010Hyd, where Xaa is any amino acid, Yaa and Zaa are small residues other than glycine and Hyd is a bulky hydrophobic residue (Phe, Leu). The most effective substrate, Arg\u2010Pro\u2010Arg\u2010Thr\u2010Ser\u2010Ser\u2010Phe, was phosphorylated with a <jats:italic>K<\/jats:italic>\n<jats:sub>m<\/jats:sub> of 5 \u03bcM and V<jats:sub>max<\/jats:sub> of 260 U\/mg. PKB\u03b1 phosphorylated histone H2B (<jats:italic>K<\/jats:italic>\n<jats:sub>m<\/jats:sub> 5 \u03bcM, <jats:italic>V<\/jats:italic>\n<jats:sub>max<\/jats:sub> 68 Ulmg) specifically at Ser\u201036 which also lies in an Arg\u2010Xaa\u2010Arg\u2010Xaa\u2010Xaa\u2010Ser\u2010Hyd motif.
