The bacterially expressed ST domain transfers the 5\u2032\u2010sulfuryl group of 3\u2032\u2010phosphoadenosine\u20105\u2032\u2010phosphosulfate (PAPS) to only deacetylated heparin with an efficiency similar to that previously reported for the purified rat HSNST. Moreover, the <jats:italic>K<\/jats:italic>\n<jats:sub>m<\/jats:sub>\n<jats:sub>PAPS<\/jats:sub> (2.1 \u03bcM) of the ST domain is also similar to that of the rat enzyme. Lys<jats:sup>48<\/jats:sup> is a key residue in mEST catalysis. The residue corresponding to Lys<jats:sup>48<\/jats:sup> is conserved in all known heparan sulfate sulfotransferases (Lys<jats:sup>614<\/jats:sup> in the ST domain of hHSNST). Mutation of Lys<jats:sup>614<\/jats:sup> to Ala abolishes <jats:italic>N<\/jats:italic>\u2010sulfotransferase activity, indicating an important catalytic role of Lys<jats:sup>614<\/jats:sup> in the ST domain.
